What is a PCR?

The PCR is a test which enables us to confirm or dismiss infection with COVID-19.

Once the patient’s swab has been taken, the first step is to extract the virus RNA from the sample and purify it. Using a variant of standard PCR, RT-PCR, we are able to convert RNA into DNA, amplify the genetic code of the virus and identify it.

PCR results

The results we get from a PCR for COVID-19 are:

  • The test has detected RNA of the virus, the result is positive. In this case, we would know that the patient has Covid-19.
  • If the genetic material of the virus is not detected, the person is not infected.

How is the PCR sample obtained?

The respiratory sample is taken from the nose and pharynx with a swab, which is then analysed in the microbiology laboratory where viral RNA extraction and subsequent PCR are performed to determine the presence of the virus. Specifically, the following steps are followed:

  • Naso-pharyngeal swabbing.
  • The patient’s head is tilted back 45º
  • The swab is gently introduced through the nose until it reaches the nasopharyngeal area
  • Once here, the swab is rotated several times and removed
  • The swab is closed in a tube, labelled with the patient’s data and analysed.

The high sensitivity of the test

The diagnostic sensitivity and specificity of the technique has been determined on a panel of anonymised positive and negative samples from the Biobank of the Centro Nacional de Microbiología (Instituto Carlos III). The analysis of the results showed a sensitivity of 100% and a specificity of 100%.
